The Power of High Content, High-Throughput Screening in Condensate-Based Drug Discovery
Biomolecular condensates are reversible, membrane-less organelles with unique compositions that regulate diverse cellular processes by selectively phase separating and concentrating biomolecules within specific regions of cells. Misregulation of condensates (a condensatopathy) has been implicated in many diseases such as cancer, neurodegeneration, and metabolic and infectious diseases. Dewpoint Therapeutics is developing small molecules that modulate condensates (c-Mods) to treat disease. To discover c-Mods, we have taken an entirely phenotypic screening approach to perform high content screens up to 400,000 compounds, directly measuring condensate function within disease relevant cells. This approach has posed challenges in terms of balancing high content imaging with the appropriate resolution to be capable of measuring very small condensates within cells and achieving the desired throughput. Herein, we describe how we have managed to address these challenges and successfully run high throughput, high content assays at scale to discover c-Mods using our custom-designed robotic platforms and generative AI.
